IncuCyte® NucLight Rapid Red Reagent for live-cell nuclear labeling

Incucyte® Nuclight Rapid Red Dye for live-cell nuclear labeling

Supplied as a single vial (50 µL), sufficient for 100 to 200 tests in a 96-well plate.

(Cat. No. 4717)

The Incucyte® Nuclight Rapid Red Dye for nuclear labeling is a cell permeable DNA stain that specifically stains nuclei in cells and is ideally suited to the mix-and-read, real-time quantification using the Incucyte® Live-Cell Analysis System. Addition of the Incucyte® Nuclight Rapid Red Dye to normal healthy cells is non-perturbing to cell growth and morphology and provides homogenous staining of nuclei, enabling the real-time, fluorescent quantification of cell proliferation. When added to tissue culture medium, the inert stain crosses the cell membrane and has excellent specificity for DNA without the need for a wash step. With the Incucyte® integrated analysis software, fluorescent objects can be quantified and background fluorescence minimized. This reagent can be multiplexed with Incucyte® Annexin V, Caspase-3/7 or Cytotox Dyes for simultaneous readouts of apoptosis or cytotoxicity alongside cell proliferation.

Incucyte® Nuclight Rapid Red Dye Quick Guide

Nuclight Rapid Red Dye is non-perturbing and enables high-efficiency labeling of cell nuclei

Optimized dye concentrations do not perturb cell health. Nuclight Rapid Red labeled HT-1080 (adherent; left graph) and Jurkats (non-adherent; right graph) cells show equivalent cell growth profiles to non-labelled cells.

Rapid, high efficiency labeling of adherent and non-adherent cell types.  After addition of Nuclight Rapid Red Dye, all cells  in the population are labeled immediately and homogenously. Note the homogeneous labeling of cell nuclei and the healthy cell morphology. HT-1080 cells (left image) and Jurkats (right image) show representative nuclear labeling and red fluorescence.

Multiplex with cell health reagents

Analysis of target tumor cell viability over 48 hours multiplexed with an apoptosis readout. HT-1080 cells were labeled with Nuclight Rapid Red Dye and cultured in the presence of Incucyte® Caspase-3/7 Green Dye. Cell death (apoptosis) is monitored in real time by measuring the loss of red object area (left) and increase of green object count (Caspase-3/7 signal) over time.

Visualize changes in the phenotype of labeled immune cells. Nuclight Rapid Red labeled human PBMC’s activated with anti-CD3 antibody and IL-2 induces T-cell aggregation or “clustering”. Images shown at are at 0, 60, and 96hrs. User friendly Incucyte® integrated software  enables accurate quantification of cell clustering over time.
